【摘要】:1.1Thisguidecoversthegeneraltechniqueof"spiking"abroadrangeofmaterialsintoaqueousmedia.Thisguidewillservetheanalystinpreparingspikedsamplesforqualitycontrolpurposes.Guidanceisalsoprovidedtoaidtheanalystincalculatingrecoveriesandinterpretingresults.Itistheresponsibilityoftheanalysttodeterminewhethertheproceduresandmaterialsdescribedhereareappropriatetothetaskathand.1.2Theproceduresinthisguidearefocusedon"matrixspike"preparation,analysis,andinterpretationofresults.Theapplicabilityoftheseprocedurestothepreparationofcalibrationstandards,calibrationcheckstandards,laboratorycontrolstandards,referencematerials,andotherqualitycontrolmaterialsbyspikingisincidental.Asample(thematrix)isfortified(spiked)withtheanalyteofinterestforavarietyofanalyticalandqualitycontrolpurposes.Whilethespikingofmultiplesampleportionsisdiscussed,themethodofstandardadditionsisnotcovered.1.3Thisguideisintendedforuseinconjunctionwiththeindividualanalyticaltestmethodthatprovidesproceduresforanalysisoftheanalyteorcomponentofinterest.Thetestmethodisusedtodetermineananalyteorcomponent''sbackgroundleveland,againafterspiking,itsnowelevatedlevel.Eachtestmethodtypicallyprovidesproceduresnotonlyforsamples,butalsoforcalibrationstandardsoranalyticalcontrolsolutions,orboth.Theseproceduresincludepreparation,handling,storage,preservation,andanalysistechniques.Theseproceduresareapplicablebyextension,usingtheanalyst''sjudgementonacase-by-casebasis,tospikingsolutions,andarenotreiteratedinthisguide.SeealsoPracticeE200forpreparationandstorageinformation.1.4Theseproceduresapplyonlytoanalytesthataresolubleinwaterattheconcentrationofthespikeplusanybackgroundmaterial,ortoanalytessolubleinasolventthatisitselfwater-soluble.Thesystemusedinthelatercasemustresultinahomogeneoussolutionofanalyteandsample.Meaningfulrecoverydatacannotbeobtainedifanaqueoussolutionorhomogenoussuspensionoftheanalyteofinterestinthesamplecannotbeattained.Theseproceduresmaybeapplicabletomicrobiologicalpreparationsifthehomogeneityofthesuspensioncanbeadequatelymaintainedthroughoutthecourseoftheanalysis,forexample,bymechanicalagitationorstirring.1.5Matrixspikingmaybeperformedinthefieldorinthelaboratory,dependingonwhichpartoftheanalyticalprocessistobetested.Fieldspikingteststherecoveryoftheoverallprocess,includingpreservationandshippingofthesample.Laboratoryspikingteststhelaboratoryprocessonly.Spikingofsampleextracts,concentrates,ordilutionswilltestonlythatportionoftheprocesssubsequenttoadditionofthespike.1.6ThevaluesstatedinSIunitsaretoberegardedasthestandard.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
